Most of the members of the Winchester family were or are hunters, but not all of them. John Winchester is the head of the family. The Winchester bloodline goes back to Cain and Abel making them the Winchesters' ancestors. Mary Winchester (née Campbell) comes from a family hunters.

Known members

  • John Winchester (deceased) - Head of the Winchester family. He is the husband of Mary. He had two sons with Mary, Dean and Sam. He had no knowledge of the supernatural world before his wife's murder. He became a hunter so he could avenge Mary. Sometime after his wife's murder, he had another son out of wedlock, Adam. He traded his life along with the Colt so Azazel would heal and resurrect Dean when he was in a catatonic state after a car crash
  • Mary Winchester (deceased) - She is the wife of John and the mother of Dean and Sam. She named her two sons after her deceased parents, Samuel and Deanna. Her parents raised her as a hunter, but she gave up hunting after she met John and both her parents were killed by Azazel. She was killed by Azazel when Sam was six months old and she went into his room to check on him and witnessed Azazel feeding baby Sam his demon blood. She never told John about the supernatural world or her former life as a hunter. Dean and Sam didn't learn about her hunter origins until Castiel sent Dean back in time, where he met younger versions of John and Mary in "In The Beginning"
  • Dean Winchester - He is the eldest son of John and Mary and the older brother of Sam and Adam. He is one of the three surviving members of the Winchester family
  • Sam Winchester - He is the youngest son of John and Mary. He is the younger brother of Dean and the older half-brother of Adam. He is one of the three surviving members of the Winchester family
  • Samuel Campbell - He is the husband of Deanna. He is the father of Mary and father-in-law of John. He is Dean and Sam's maternal grandfather. His grandson, Sam, was named after him. Deanna and he were both hunters and raised Mary to be one as well. He was killed by Azazel. He was brought back to life along with Sam at the end of fifth season, but no one knows who did it yet or why.
  • Deanna Campbell (deceased) - She is the wife of Samuel. She is the mother of Mary and mother-in-law of John. She is the maternal grandmother of Dean and Sam. Her grandson, Dean, was named after her. She and Samuel were both hunters and raised Mary to be one as well. She was killed by Azazel
  • Adam Milligan - He is the youngest son of John. He is one of the three surviving members of the Winchester family. He was conceived sometime during John's time as a hunter. He is the younger half-brother of Dean and Sam. Unlike his brothers, John did not tell him about the supernatural world. John and he did not have a strong relationship, but when John visited him they would do normal father-son activities. He and his mother were killed by ghouls that were the children of a ghoul John had killed. He never met his half-brothers and John most likely never told him about them due to the fact that Dean and Sam never knew of his existence until the ghoul disguised as Adam called them. Regardless of not being a hunter, his half-brothers gave him a hunter's funeral on the reasoning that Adam died the death of a hunter so he deserved the funeral of one. Eventually, he was brought back to life by the angels under the impression that he would be the vessel of Michael, but really bait to get Dean to say "Yes" to Michael. When this plan failed, Michael gave up on Dean and decided to use Adam as a vessel. He is still currently being possessed by Michael and alive in Lucifer's Cage
  • Christian Campbell - He is Sam and Dean's third cousin. He is a calm and skilled Hunter working with Samuel Campbell, his grandfather.
  • Gwen Campbell - She is Sam and Dean's third cousin. She is a Hunter working with Samuel Campbell, her grandfather.
  • Mark Campbell - He is Sam and Dean's distant cousin. He is a Hunter working with Samuel Campbell, his grandfather.
  • Unnamed Uncle/Brother (Mary's brother or uncle) - Unknown if he a hunter or not, or if he alive. Also unknown if he had any children.
  • Cain and Abel (deceased) - It was revealed by the Archangel Michael that through the Winchester bloodline, Dean and John are descendants of Cain and Abel, making John a temporary vessel and Dean his true vessel. And since Sam and Adam are John's biological sons that makes Cain and Abel their ancestors as well

Allies

  • Bobby Singer - A fellow hunter, who helps the Winchester brothers with many of their cases. He also sometimes sends the boys to places that he believes are experiencing the supernatural. The brothers see Bobby as a surrogate father. When they were children, Dean and Sam would call him "Uncle Bobby"

  • Ellen Harvelle (deceased) - A fellow hunter, who was a friend of John Winchester. Like Bobby, she gave guidance to Sam and Dean about hunting. She did like a mother-figure to the two brothers, lecturing them about not calling her enough to inform her that their alright. She died trying to help Dean and Sam kill Lucifer with the Colt
  • Jo Harvelle (deceased) - A fellow hunter, who was a friend to Dean and Sam. Although nothing serious happened between the two, she and Dean were attracted to and harbored romantic feelings for one another. She died trying to help Dean and Sam kill Lucifer with the Colt
  • Ash (deceased) - A friend to Dean and Sam. Although not a hunter, he attempted to help the brothers track down Azazel
  • Castiel - The angel that pulled Dean out of Hell and resurrected him under God's command. He was looked down upon and demoted by his angel brethren due to him getting close to Dean and actually caring for him. He rebelled against the angels by trying to help Dean stop the Apocalypse. When Lucifer was released, he helped the Winchester brothers try to kill him and when that failed, he helped the brothers trap Lucifer. During his rebellion, he showed his loyalty to the Winchester brothers by killing several of angelic siblings
  • Anna Milton (formerly, deceased) - The Winchester brothers protected Anna from demons and angels after they learned that she had the ability to listen to angels's conversations. They eventually helped her regain her memories of her past life as an angel. During their time together, Anna and Dean became very close to one another to the point where they became intimate. Anna attempted to get Dean to forgive himself for torturing souls in his time in Hell. Ultimately, she betrayed the Winchesters after Lucifer was freed. She went back in time to 1978 in an attempt to kill John and Mary Winchester before either brother was born, which would cause Dean and Sam to never exist
  • Pamela Barnes (deceased) - A psychic, who was a friend of Bobby Singer. She lost her eyes trying to figure out what being rescued Dean's soul from Hell and brought him back to life, only learning the being's name, Castiel. She also helped the brothers help Anna in regaining her memory as an angel. She returned again to use her powers to turn the brothers into spirits in a town where the local Reaper was captured and no one could die. She lost her life protecting the brothers' bodies from demons when the brothers' souls were still out of their bodies
  • Ruby (formerly, deceased) - Ruby became Sam's mentor on how to use his powers so that he had the power to kill Lilith. When Dean was dead, the two of them became very close and were intimate. Ultimately, she revealed that her true loyalty was to Lucifer and that she trained Sam so that he could kill Lilith and break the final seal. Regardless of her loyalty to Lucifer, Ruby did help the Winchester brothers with many of their cases, such as helping to save Anna from being captured by demons which would have helped her side. She also provided the brothers with their two most powerful weapons, her knife that has the power to kill demons and the Colt, which she helped Bobby fix after all the original bullets for it were used
  • Chuck Shurley (God) - A prophet that sees the future of the Winchester brothers. He would use his visions of the future to help the brothers with matters of the Apocalypse. In "Swan Song", the show gives the impression that Chuck is the alter ego of God if that's so that would mean He is also the brothers' creator
